柠檬酸盐合酶是三羧酸（TCA）循环的初始酶。该酶催化乙酰辅meiA（乙酰辅meiA）与草酰乙酸之间的反应，形成柠檬酸和具有硫醇基团（CoA-SH）的辅meiA。该
比色测定基于5'，5'-二硫代双2-硝基苯甲酸（DTNB）和CoA-SH之间的反应以形成TNB，其在412nm处表现出最大吸光度。
吸光度的强度与柠檬酸盐合酶活性成正比。这种酶是线粒体基质的标志物。
